I have written about the bus service from various points in the city to the new airport at Shamsabad. In that post I provided a map of the Begumpet area with the Paryatak Bhavan highlighted. Since then some more information has cropped up.
The buses run every 30 minutes from Paryatak Bhavan, Begumpet, and the run takes about 1 hr 30 mins. No stops in between. There is likely to be a rush for seats. Tickets are available on the bus. The fare is Rs. 95/-, inclusive of taxes.
The second boarding point is Sarojini Devi Eye Hospital at Mehdipatnam. The hospital is actually in Humayun Nagar on the road from Masab Tank (NMDC Office) to Mehdipatnam. See maps below.

At this time I have no information about the exact spot near Sarojini Devi Eye Hospital where the shuttles pick up passengers. I do know that the area is crowded with shops and sees a lot of traffic. The road from Masab Tank towards Mehdipatnam is the main conduit for all traffic going towards the Old Bombay Highway, Bangalore Highway & the Srisailam Highway. Plus the area is densely populated.
If you are traveling from the Madhapur area it might be better to board the shuttle from Begumpet.

The airport shuttle route from begumpet to Airport is via punjagutta- masab tank – medhipatnam – aramghar – airport
and if u board the bus at Sarojini Devi Eye Hospital you can avoid traveling via punjagutta (twice infact if you go from madhapur to begumpet to board the shuttle) – masab tank and on any day I would assume that going to medhipatnam is much easier than going to begumpet from madhapur specially with the new road near raheja mindspace to gachibowli

You are right about having to cross Panjagutta twice. My contention is that the Sarojini Devi Eye Hospital area is very congested and the boarding point would most probably be by the road side.
The Tourism Plaza in Begumpet would provide a more comfortable boarding point. Secondly there are more commute options available from Madhapur to Begumpet. The Begumpet railway station is less than 1/2 km from the Tourism Plaza and MMTS can be a good option when traveling from Madhapur. Provided of course that you don’t have too much luggage.
